You can't use it on stage1 as you need to actually deal damage first, so doesn't work on this buggy test phase

For example in pka say you do 10m damage to boss on stage4, that boss has 1mil HP. You did 9mil overkill

On stage5 you can then unleash doffys special and do 9mx2=18mil dmg

Good old V2 Doffy. The reason for the normal attacks only debuff.

You have to beat an enemy and the overkill damage is what's calculated in the special. It says you don't have any so the special won't launch

A Enemy has 4M Health you deal 8M damage. The 4M overdamage will be doubled by this Special.
The SP Base: 0 will tell you what over damage you have dealt

No, it's only damage dealt beyond the enemies hp, for example an enemy has 50k HP and you do 100k damage, the extra 50k damage you did goes to doffys special
